Notch 1 Antibody (C-10) is a mouse monoclonal IgG2b kappa light chain antibody developed to detect human Notch1, also known as Neurogenic locus notch homolog protein 1. Notch1 monoclonal antibody (C-10) targets an epitope between amino acids 2433-2467 near the C-terminus of Notch1, ensuring precise recognition of the Notch1 precursor, mature Notch1, Notch1 NEXT, and Notch1 NICD forms across multiple species, including mouse, rat, human, and equine, in applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Notch1 is a transmembrane receptor integral to cell fate determination during development and tissue homeostasis. Post-translational modifications (PTMs) of Notch1, such as glycosylation, phosphorylation, and ubiquitination, are crucial for function. Glycosylation of the extracellular domain influences ligand binding and receptor activation, while phosphorylation and ubiquitination of the intracellular domain regulate stability and degradation, thereby modulating signal transduction. Dysregulation of these PTMs can lead to aberrant Notch1 signaling, contributing to various pathologies, including cancers and developmental disorders. Anti-Notch1 antibody (C-10) is available in non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, catering to diverse experimental needs. I tested it on protein extracts (30ug) of a cell line of trophoblasts induced to differentiate to an endothelial-like phenotype.
