NIK Antibody (A-12) is a mouse monoclonal IgG2b kappa light chain antibody designed to target the NFκB Inducing Kinase (NIK), also known as MAP3K14, across human, mouse, and rat species. NIK monoclonal antibody (A-12) specifically recognizes an epitope within amino acids 700-9473 at the C-terminus of the NIK protein, ensuring precise detection in various applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). NIK is a crucial serine/threonine kinase involved in the non-canonical NFκB signaling pathway, where NIK plays a significant role in the activation and processing of NFκB transcription factors. These transcription factors are essential for regulating immune responses, inflammation, and cell survival, making NIK a key player in maintaining immune homeostasis. By interacting with and acting as a co-stimulator for IkB kinase α (IKKα), NIK phosphorylates IkB, leading to its degradation and the subsequent activation of NFκB. Dysregulation of NIK has been implicated in various autoimmune diseases and cancers, highlighting NIK′s importance in disease pathology. Can NIK (A-12): sc-8417 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, NIK (A-12): sc-8417 has been validated for use in IHC with paraffin-embedded sections. Rated 1 out of 5 by from Antibody does not work in Western blot20µg total lysate were loaded from gastric cancer epithelial cells untreated and treated with 20µM MG-132 for 4 hours. Many unspecific bands were observed, none corresponded to NIK, which should increase in the MG-132-treated cells.
