NFATc3 Antibody (F-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects NFATc3 of mouse, rat, and human origin through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry (IHC), and enzyme-linked immunosorbent assay (ELISA). NFATc3 monoclonal antibody (F-1) is offered in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and various Alexa Fluor® conjugates. NFATc3, also known as nuclear factor of activated T cells 3, plays a crucial role in the immune response by regulating the expression of cytokines in T cells. NFATc3 is part of the NFAT family of transcription factors, which are activated by increases in intracellular calcium levels. This activation leads to the dephosphorylation of NFAT proteins by the calmodulin-dependent phosphatase calcineurin, resulting in a conformational change that exposes the nuclear localization signal. This change is vital as NFAT proteins translocate from the cytosol into the nucleus, where these proteins bind to DNA and modulate gene expression. NFATc3 interaction with other proteins, such as AP-1 proteins (Fos and Jun), further underscores NFATc3′s importance in forming cooperative complexes that regulate immune responses.

I would like to use sc-8405 (mouse anti-NFATc3) in IHC. Which antigen retrieval buffer was used?

Thank you for your question. We recommend using sodium citrate buffer (pH 6.0) with heat treatment for antigen retrieval.

If I want to use SC-8405 conjugated with FITC in mice tissue immunofluorescence staining, what buffer should I use? Do I need to add blocking buffer?

Thank you for your question. For direct immunofluorescence staining with the FITC conjugated primary antibody (sc-8405 FITC), we recommend first blocking for 30 minutes with 10% normal blocking serum in PBS or in UltraCruz® Blocking Reagent (sc-516214). If blocking serum is used, it should ideally be derived from the same species in which the primary antibody was raised. Wash with three changes of PBS for 5 minutes each. Incubate with primary antibody conjugated to a fluorophore for 90 minutes at room temperature, or overnight at 4° C. Optimal antibody concentration should be determined by titration; recommended range is 4–10 µg/ml in either PBS with 1.5% normal blocking serum or in UltraCruz® Blocking Reagent (sc-516214). https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

We recommend using a directly conjugated mouse monoclonal primary antibodies. This product, sc-8405, is available directly conjugated to AF488, AF647, and FITC.
