Myotrophin Antibody (A-10) is a mouse monoclonal IgG2a antibody that detects Myotrophin in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Myotrophin is a crucial cytoplasmic protein that plays a significant role in cellular signaling, particularly during sustained activation of the nuclear factor kappa-light-chain-enhancer of activated B cells (NFκB). Myotrophin translocates to the nucleus, where Myotrophin can influence gene expression and cellular responses. Myotrophin is essential for the differentiation of cerebellar neurons, especially granule cells, and is involved in cerebellar morphogenesis. Structurally, Myotrophin consists of 117 amino acids, featuring an acetylated N-terminus and two internal ankyrin repeats, which are important for protein-protein interactions and stability. Myotrophin has been linked to cardiac hypertrophy, as Myotrophin can induce the expression of protooncogenes such as ANF and β-Myosin heavy chain, particularly in response to stimuli like cyclic stretch or inflammatory cytokines such as tumor necrosis factor α (TNFα) and interleukin-1β. Notably, Myotrophin is predominantly expressed in the brain, with significantly lower levels found in skeletal muscle, highlighting its specialized role in neuronal function and development.
