Myosin VI Antibody (G-6) is a mouse monoclonal IgG1 antibody that detects Myosin VI in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Myosin VI is a unique molecular motor that plays a critical role in intracellular vesicle and organelle transport, distinguished by binding to the pointed end of actin filaments, a feature not shared by other myosins. Myosin VI is essential for endocytosis, associating with clathrin-coated vesicles and interacting with proteins like disabled 2, highlighting importance in cellular trafficking processes. Myosin VI is predominantly expressed in the sensory hair cells of the cochlea, where maintaining the structural integrity of stereocilia bundles is crucial; defects can lead to autosomal dominant nonsyndromic sensorineural deafness in humans. The human Myosin VI gene is located in the centromeric region of chromosome 6, showing syntenic homology with mouse chromosome 9, where the Snell′s Waltzer mutation is found. This mutation results in significant auditory and behavioral impairments, including hyperactivity and lack of responsiveness to sound, due to stereocilia disorganization in the inner ear. Anti-Myosin VI antibody (G-6) serves as an invaluable tool for researchers studying molecular mechanisms underlying hearing loss and cellular dynamics of vesicle transport.
