Myosin VI Antibody (A-9) is a mouse monoclonal IgG3 kappa light chain antibody that detects Myosin VI (MYO6) of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Myosin VI antibody (A-9) is available as a non-conjugated format. Myosin VI (MYO6) plays a crucial role as a molecular motor in intracellular vesicle and organelle transport, uniquely binding to the pointed end of actin filaments, which is essential for cellular processes. MYO6 has only one light chain in its lever-arm domain, resulting in highly irregular stepping patterns and diverse step sizes compared to other myosins. Myosin VI (MYO6) associates with Clathrin-coated vesicles and disabled 2, highlighting significant involvement in endocytosis. Mouse MYO6 is predominantly expressed in the sensory hair cells of the cochlea, where MYO6 is vital for auditory function. Human MYO6 is mapped to the centromeric region of chromosome 6, which shares syntenic homology with the corresponding region on mouse chromosome 9, where the Snell′s Waltzer mutation is located. This mutation leads to behavioral effects such as lack of responsiveness to sound, hyperactivity, head tossing, and circling, attributed to the disorganization and fusion of stereocilia bundles in the inner ear. Defects in MYO6 are linked to autosomal dominant nonsyndromic sensorineural deafness in humans, underscoring MYO6′s critical role in hearing. Human MYO6 is expressed in the fetal cochlea and brain, as well as in the adult brain, emphasizing MYO6′s importance in both developmental and adult physiology.
