MMP-1/8 Antibody (A-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects matrix metalloproteinases MMP-1 and MMP-8 across human, mouse, and rat species by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Raised against amino acids 100-399 within an internal region of human MMP-1, MMP-1/8 (A-7) antibody recognizes MMP-1, also known as interstitial collagenase, fibroblast collagenase, or collagenase-1, and MMP-8, also known as neutrophil collagenase or collagenase-2. Both MMP-1 and MMP-8 are enzymes critical for extracellular matrix remodeling, degrading components like collagen, gelatin, and fibronectin. Structurally, these proteins contain a pro-peptide domain, a catalytic domain with a zinc-binding site essential for proteolytic activity, and a hemopexin-like domain that influences substrate specificity and interaction with tissue inhibitors of metalloproteinases (TIMPs). The zinc-binding catalytic domain facilitates collagen fiber cleavage, a process vital in normal tissue remodeling but detrimental when unregulated. Abnormal expression of MMP-1 and MMP-8 is associated with pathological conditions such as rheumatoid arthritis and periodontal disease, where these enzymes contribute to tissue degradation.
