MLH1 Antibody (B-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects MLH1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MLH1 (B-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MLH1 plays a crucial role in the DNA mismatch repair (MMR) pathway, which maintains genetic stability by correcting errors during DNA replication. Deficiencies in this pathway, often due to mutations or epigenetic silencing of MMR genes like MLH1, are linked to various cancers, including hereditary nonpolyposis colorectal cancer (HNPCC), where pathogenic mutations in MLH1 significantly increase the risk of colorectal and other cancers. MLH1 is expressed in various tissues, including lymphocytes, heart, colon, breast, lung, spleen, testis, prostate, thyroid, and gall bladder, and MLH1 expression is often lost in several cancer types, leading to microsatellite instability and a mutated phenotype. Notably, certain cancer cell lines, such as leukemia CCRF-CEM and colon HCT 116, exhibit complete deficiency of MLH1, while MLH1 is expressed in a significant percentage of melanomas and squamous cell carcinomas. anti-MLH1 antibody (B-12) detection capabilities make this reagent an invaluable tool for researchers studying cancer development mechanisms and DNA repair pathways in tumorigenesis.

I would like to map MLH1 sites in the pig genome by ChIP-Seq in pig (Sus scrofa) spermatocytes and wonder whether this antibody will be suitable for this technology and species. Thanks in advance

Anti-MLH1 Antibody (B-12): sc-271978 is recommended for detection of MLH1 of mouse, rat, human and origin by WB, IP, IF and ELISA; also reactive with additional species, including and canine, bovine, porcine and avian. It has not been tested for ChIP-Seq applications in particular, but the agarose conjugate sc-271978 AC is suitable for pulldown experiments.

I tested it for immunofluorescence staining on mouse spermatocyte surface spread and expected for a focus staining on crossover site.
