MGMT Antibody (MT5.1) is a mouse monoclonal IgG1 kappa light chain antibody raised against recombinant MGMT of human origin, specifically reactive to MGMT in human samples by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. MGMT, also known as O6-methylguanine-DNA methyltransferase or AGT (O-6-alkylguanine DNA alkyltransferase), is a crucial DNA repair protein that removes alkyl groups from the O6 position of guanine, thereby protecting the genome from mutations caused by alkylating agents such as carcinogens and cytostatic drugs. This function is vital for maintaining genomic stability and preventing carcinogenesis, as O6-alkylguanine lesions can lead to mispairing during DNA replication, resulting in mutations. MGMT expression is transcriptionally activated in response to DNA damage, but induction by ionizing radiation requires functional p53, a tumor suppressor protein. Studies have shown that MGMT is inducible only in cells expressing wild-type p53, while cells lacking functional p53 do not exhibit this induction, suggesting that p53 plays a regulatory role. High MGMT activity has been associated with the presence of mutant p53 in certain cancer cell lines, indicating that wild-type p53 may down-regulate basal MGMT promoter activity. Understanding MGMT expression levels is important in cancer research and therapy, as elevated MGMT can confer resistance to alkylating chemotherapeutic agents, making anti-MGMT antibody (MT5.1) a valuable tool for researchers studying DNA repair mechanisms and the p53 regulatory pathway.

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, MGMT (MT5.1): sc-33674 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
