MGMT Antibody (E-1) is a mouse monoclonal IgG2a kappa light chain antibody specific for an epitope mapping between amino acids 9-24 at the N-terminus of human MGMT, showing reactivity with MGMT from mouse, rat, and human origins by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). MGMT, also known as O6-methylguanine-DNA methyltransferase or O-6-alkylguanine DNA alkyltransferase (AGT), is a vital DNA repair enzyme that removes mutagenic and cytotoxic alkyl groups from the O6 position of guanine in DNA. Such alkylation lesions can lead to mispairing during DNA replication, resulting in mutations and potentially carcinogenesis. MGMT is predominantly located in the cell nucleus, where direct access to genomic DNA allows for efficient repair of DNA damage before cell division occurs. Nuclear localization ensures maintenance of genomic stability and prevents propagation of DNA errors to daughter cells. High levels of MGMT can confer resistance to alkylating chemotherapeutic agents, making MGMT expression significant in cancer research and therapy development. Anti-MGMT antibody (E-1) serves as a valuable tool for researchers investigating DNA repair mechanisms and MGMT expression in cancer treatment outcomes.
