MDM2 Antibody (D-7) is a mouse monoclonal IgG2b kappa light chain antibody raised against amino acids 100-320 of human MDM2, exhibiting reactivity with mouse, rat, and human samples. MDM2, also called Murine Double Minute 2, is an oncoprotein that serves as a critical negative regulator of the tumor suppressor p53, playing a pivotal role in cell cycle control and apoptosis. Structurally, MDM2 comprises several key domains: an N-terminal p53-binding domain, a central acidic domain, a zinc finger domain, and a C-terminal RING finger domain endowed with E3 ubiquitin ligase activity. Anti-MDM2 monoclonal antibody (D-7) targets the central region of MDM2, including the acidic domain, which is essential for its regulatory interactions. MDM2 binds to the transactivation domain of p53 via its N-terminal domain, inhibiting p53′s transcriptional activity and promoting ubiquitination and subsequent proteasomal degradation through the RING finger domain. This interaction is crucial for maintaining appropriate levels of p53 under normal physiological conditions. However, overexpression or amplification of the MDM2 gene can lead to excessive degradation of p53, impairing p53′s ability to induce cell cycle arrest or apoptosis in response to DNA damage, and thereby contributing to tumorigenesis. Understanding MDM2 structure provides insights into regulatory mechanisms and potential therapeutic targets, especially in cancers where p53 is wild-type but p53 function is inhibited by MDM2 overactivity. MDM2 (D-7) antibody is suitable for applications such as western blotting, immunoprecipitation, immunofluorescence, enzyme-linked immunosorbent assays, and immunohistochemistry, making MDM2 (D-7) antibody a valuable tool for researchers investigating MDM2 expression, structure-function relationships, and role in cancer development.

Can MDM2 (D-7): sc-13161 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, MDM2 (D-7): sc-13161 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
