MCM2 Antibody (E-8) is a mouse monoclonal IgG2b kappa light chain antibody that detects MCM2 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MCM2 (E-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The mini-chromosome maintenance (MCM) family of proteins, which includes MCM2, plays a crucial role in the regulation of DNA replication, ensuring that DNA is replicated only once during each cell cycle. MCM2 is particularly important as MCM2 forms part of the MCM2-7 complex, which acts as a helicase to unwind DNA, a critical step for the initiation of DNA replication. This unwinding is essential for proper genome duplication, and any dysregulation can lead to genomic instability, a hallmark of many cancers. MCM2 expression increases during cell growth, peaking at the transition from G1 to S phase of the cell cycle, and is tightly regulated by E2F transcription factors and various protein kinases. Additionally, MCM2/MCM4 complexes serve as substrates for Cdc2/cyclin B, highlighting MCM2′s role in cell cycle regulation. MCM3 cleavage during apoptosis, which can be inhibited by caspase inhibitors, leads to MCM complex inactivation, further emphasizing MCM proteins′ importance in maintaining cellular integrity and function.

Verified specificity with blocking peptide and non dividing resting cells. 647 pairs well with 594 costaining with two different ABs. The typical bleedthrough of fluorescence 647 to 594 is 0, and 594 to 647 channel is only 5-10%. Excellent AB overall.

Rated 3 out of 5 by from MCM2 AbAntibody works for WB. But signal is quite weak. Protein was weakly detected at dilution 1:100 using HCT116 whole cell lysates. Band corresponding to MCM2 protein was observed around 110-120kDa.
