Keap1 Antibody (G-2) is a mouse monoclonal IgG2b kappa light chain antibody that detects Keap1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Keap1 antibody (G-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Keap1, also known as kelch-like ECH-associated protein 1, plays a crucial role as a stress-sensing adaptor in the Cullin3 (Cul3)-dependent E3 ubiquitin ligase complex, which is essential for the regulation of Nrf2 (NF-E2-related factor 2). This regulation is vital because Nrf2 is a key transcription factor that mediates the expression of cytoprotective genes in response to oxidative stress and electrophilic insults. Under normal conditions, Keap1 maintains low levels of Nrf2 by promoting ubiquitination and subsequent degradation, thereby preventing excessive activation of protective genes. During oxidative stress, modifications to specific cysteine residues in Keap1, particularly Cys 273 and Cys 288, lead to a conformational change that releases Nrf2, allowing nuclear translocation and target gene activation. This dynamic interplay between Keap1 and Nrf2 is critical for cellular defense mechanisms, making Keap1 an important protein in the study of oxidative stress responses and potential therapeutic targets for diseases related to oxidative damage.
