Keap1 Activators

Kelch Like ECH Associated Protein 1 (Keap1) is a cysteine-rich protein that plays a crucial role as a substrate adaptor protein for the Cullin3 (Cul3)-based E3 ubiquitin ligase complex. This complex targets nuclear factor (erythroid-derived 2)-like 2 (Nrf2) for ubiquitination and subsequent degradation. Nrf2 is a critical transcription factor that regulates a host of antioxidant and detoxification genes. Keap1 is primarily activated through the modification of its cysteine residues. Chemicals such as Sulforaphane, Curcumin, and CDDO-Im can directly modify these cysteine residues in Keap1, thereby disrupting its ability to bind to Nrf2. This releases Nrf2 to move into the nucleus, where it can induce the transcription of genes involved in cellular defense mechanisms. Other chemicals like Zinc and Luteolin can bind to Keap1, leading to a conformational change that disrupts its interaction with Nrf2.

Another method of activation includes altering the conformation of Keap1. Chemicals such as Oltipraz, Luteolin, and Quercetin bind to Keap1 and alter its structure, allowing for the release of Nrf2. Alkylation is another route through which Keap1 can be activated. Diallyl sulfide, for instance, alkylates Keap1 and leads to the release and activation of Nrf2. Oxidation is a fourth method by which Keap1 is activated. For example, tert-Butylhydroquinone (tBHQ) oxidizes specific cysteine residues in Keap1, which then leads to the disassociation with Nrf2 and its subsequent activation. These chemicals showcase a variety of mechanisms, including alkylation, oxidation, direct binding, and structural alteration, through which they can activate Keap1. These activations in turn lead to various cellular defense responses orchestrated through the Nrf2 pathway.
