JNK1 Antibody (G-4) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 358-397 at the C-terminus of human JNK1, a stress-activated protein kinase critical for cellular stress responses. JNK1 mouse monoclonal antibody (G-4) recognizes an epitope within this C-terminal region and demonstrates broad species cross-reactivity, detecting JNK1 in mouse, rat, and human samples through Western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) applications. The molecular complexity of JNK1 signaling reflects its sophisticated regulatory roles: encoded by a gene (PRKM8, SAPK1, MAPK8) located on chromosome 10q11.22, JNK1 shares 83% amino acid identity with JNK2. Through alternative mRNA splicing, JNK1 contributes to the production of ten distinct isoforms, including both p46 and p54 variants, enabling precise control of cellular responses. JNK1′s significance in disease becomes particularly evident in hepatocellular carcinoma (HCC), where JNK1′s activation status correlates with tumor progression through modulation of critical oncogenic pathways involving Jun, Fos, and Myc. Beyond cancer regulation, JNK1 orchestrates sophisticated apoptotic responses by phosphorylating p53, triggering expression of pro-apoptotic genes like BAX and PUMA. This phosphorylation cascade extends to mitochondrial regulation, where JNK1 influences membrane permeability and cytochrome C release in the intrinsic apoptotic pathway. JNK1 participates in complex stress response networks, interacting with kinases such as MEKK1 and p38. These interactions become particularly relevant in cancer contexts: capsaicin activates both JNK1 and p38 in ras-transformed breast epithelial cells, while nitrogen oxides (NOx) trigger coordinated upregulation of JNK1 with c-Fos and c-Jun.
