JNK Antibody (D-2) is a mouse monoclonal IgG1 kappa light chain antibody raised against full-length human JNK2 (amino acids 1-424), a crucial stress-activated protein kinase. JNK mouse monoclonal antibody (D-2) recognizes an epitope within the JNK immunogen sequence and demonstrates broad species cross-reactivity, detecting JNK proteins in mouse, rat, and human samples through Western blotting (WB), immunoprecipitation (IP), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA) applications. Anti-JNK monoclonal antibody (D-2) is available in non-conjugated form and multiple conjugated forms including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, offering versatility for diverse experimental needs. The JNK proteins, also known as MAPK8/9/10 or SAPK/JNK, function as serine/threonine kinases that respond to stress stimuli, cytokines, and growth factors by phosphorylating and activating transcription factors including c-Jun, ATF2, and Elk-1. Three distinct genes (JNK1, JNK2, and JNK3) produce ten different isoforms through alternative mRNA splicing, including p46 and p54 variants, each contributing to the regulation of cellular processes such as proliferation, differentiation, apoptosis, and stress responses. JNK participates in sophisticated protein networks, forming complexes with scaffold proteins like JIP (JNK-interacting proteins), Bcl-2 family members involved in apoptosis regulation, and other signaling kinases such as MEKK1 and p38 MAPK. These interactions create intricate signaling cascades that influence cell growth and transformation. When JNK signaling becomes dysregulated, the resulting disruption can contribute to various diseases including cancer, neurodegenerative disorders, diabetes, and cardiovascular conditions. Through broad reactivity with all JNK1, JNK2, and JNK3 p46 and p54 isoforms, JNK (D-2) antibody serves as a valuable tool for investigating these complex signaling networks and their roles in both normal cellular functions and disease states.
