HR Antibody (D-4) is a mouse monoclonal IgG1 antibody that detects HR (Hairless) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). HR is a 1,189 amino acid protein that exists in two isoforms produced by alternative splicing, with isoform 1 being more prevalent and expressed at low levels in the kidneys and testis, while isoform 2 is predominantly found in the skin. Both isoforms are co-expressed in various tissues, showing strong expression in the small intestine and brain, and weak expression in the trachea. HR functions as a transcription factor that plays a crucial role in hair growth regulation, which occurs in three distinct phases: anagen (growth), catagen (regression), and telogen (rest). Although the exact mechanisms remain unclear, HR (D-4) antibody targets a protein that influences one of these hair growth phases and collaborates with the vitamin D receptor (VDR) to modulate hair follicle cycling. Deficiencies in HR can lead to severe conditions such as alopecia universalis congenita (ALUNC) and atrichia with papular lesions (APL), both of which are autosomal recessive disorders. ALUNC is characterized by the absence of hair despite the presence of hair follicles, while APL is marked by the development of papular lesions across the body and minimal to no hair growth. HR (D-4) antibody serves as an essential tool for researchers investigating the molecular mechanisms underlying hair growth and related disorders.
