HR Inhibitors

The class of chemicals collectively referred to as HR Inhibitors encompasses a range of compounds that can interfere with the HR protein function indirectly by modulating associated signaling pathways, particularly the Wnt/β-catenin pathway, which is implicated in hair follicle development and, by association, in the functioning of the HR protein. These compounds act on different components of the Wnt pathway, which is known to interact with the HR protein in its role in hair growth regulation.

For instance, flavonoids like quercetin can inhibit Wnt signaling, which in turn can influence the HR protein's regulatory actions in hair growth. Tankyrase inhibitors such as XAV-939 act by stabilizing Axin, a negative regulator of the Wnt pathway, which would lead to the degradation of β-catenin, a transcriptional co-activator involved in hair follicle development. Similarly, compounds like ICG-001 and JW55 directly disrupt the interaction between β-catenin and its transcriptional partners within the Wnt pathway, which could have downstream effects on HR activity. Additional chemicals in this class, such as LGK-974, target enzymes like Porcupine that are essential for post-translational modification and secretion of Wnt proteins, thereby impacting the signaling that could modulate HR function. KY02111, FH535, PKF118-310, and IWR-1 all target various aspects of the Wnt/β-catenin signaling cascade, from ligand production to nuclear transcriptional activity, which may intersect with HR protein functionality.
