GPx-4 Antibody (E-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects GPx-4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-GPx-4 antibody (E-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GPx-4 plays a crucial role in cellular defense against oxidative stress, primarily by reducing lipid hydroperoxides to their corresponding alcohols and converting free hydrogen peroxide into water, thereby preventing lipid peroxidation of cell membranes. GPx-4 is unique among glutathione peroxidases as GPx-4 contains selenocysteine at its active site, which is essential for enzymatic function and is encoded by the stop codon TGA. This post-translational modification allows GPx-4 to maintain antioxidant activity, which is particularly important in protecting cells from oxidative damage that can lead to various diseases, including neurodegenerative disorders and cancer. GPx-4 forms high molecular weight oligomers, which may enhance stability and functional capacity in cellular environments. The diverse roles of GPx-4 in different tissues underscore significance in maintaining cellular homeostasis and responding to oxidative challenges.

Hello Which is the recomended protocol for cell fixation and permeabilization to be used in flow cytometry? And the recomended staining dilution?

Thank you for your question. For cell fixation and permeabilization, we recommend using our FCM Fixation Buffer (sc-3622) and FCM Permeabilization Buffer, sc-3623. We suggest 1 µg of fluorochrome-conjugated antibody per 100 µl of the prepared cell suspension (equal to 1 million cells). What is the difference between GPX-4 E-12 and B-12? E-12 is suitable for SHSY-5Y cells?

Thank you for your question. These antibodies have been raised against the same amino acid range but bind to different epitopes within that region. While GPx-4 Antibody (B-12) is not recommended for IHC/IHC(P) use, both antibodies are suitable for the detection of GPx-4 in any cells or tissues of mouse, rat or human origin by WB, IP, IF, and ELISA.

Hi there. I have some questions for the antibody, GPx-4 (E-12) sc-166570 . Should this antibody be made in a 5% skim milk blocking solution in PBST? Or should a different blocking solution be used? Similarly, should washing steps be performed in PBST?

Thank you for your question. We recommend using 5% nonfat milk in TBST for blocking and for primary and secondary antibody incubations. We recommend washing in TBST, with a final wash in TBS without Tween-20. Please refer to our Western blotting protocol for more detailed information.
