GPD2 Antibody (D-9) is a mouse monoclonal IgG1 antibody that detects GPD2 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). GPD2, also known as glycerol-3-phosphate dehydrogenase 2 or GDH2, is a 727 amino acid protein that plays a crucial role in cellular metabolism by catalyzing the conversion of glycerol-3-phosphate to dihydroxyacetone phosphate while reducing enzyme-bound flavin adenine dinucleotide (FAD). This enzymatic activity is vital for the glycerol phosphate shuttle, which facilitates the reoxidation of NADH produced during glycolysis, thereby linking carbohydrate metabolism to lipid metabolism. GPD2 is predominantly localized to the outer surface of the inner mitochondrial membrane, where GPD2 works in concert with GPD1, a cytosolic NAD-linked glycerol-3-phosphate dehydrogenase, to maintain cellular energy homeostasis. Notably, GPD2 is highly expressed in human pancreatic islets, where GPD2 is essential for the glucose-sensing function of pancreatic beta cells; decreased levels of GPD2 have been associated with impaired glucose-stimulated insulin release, a hallmark of noninsulin-dependent diabetes mellitus. GPD2 exists in two alternatively spliced isoforms and features two EF-hand domains, which are important for structural stability and function. GPD2 maps to human chromosome 2q24.1, highlighting GPD2′s significance in metabolic regulation and potential implications in metabolic disorders.
