GLI-3 Antibody (B-4) is a mouse monoclonal IgG2b kappa light chain antibody that detects GLI-3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). GLI-3 (B-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GLI-3, also known as GLI3FL, is a crucial regulator in the Sonic hedgehog (Shh) signaling pathway, functioning as both a transcriptional activator and repressor. This dual role is vital for proper embryonic development, particularly in the formation of limbs and craniofacial structures. GLI-3 is a member of the GLI family of zinc-finger proteins, characterized by C2H2-type zinc-finger domains that facilitate DNA binding and transcriptional regulation. GLI-3 is predominantly located in the nucleus and cytoplasm, where GLI-3 interacts with various transcriptional co-factors to modulate gene expression. Mutations in GLI-3 can lead to developmental disorders such as Greig cephalo-poly-syndactyly syndrome (GCPS), highlighting GLI-3′s significance in normal developmental processes.

When should the TransCruz reagent sc-74478 X be used instead of the standard form sc-74478?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Rated 5 out of 5 by from GLI3 IHC ReviewAntibody worked very well for IHC, nice specific nuclear staining at 1:50, used 10mins antigen retrieval with citrate buffer. Used a vectastain elite ABC kit, with DAB from a kit from R&D systems. Worked it up in placenta and also stained nicely in some tumour samples we examined.
