GLI-1 Antibody (C-1) is a mouse monoclonal IgG1 kappa light chain antibody engineered to recognize GLI-1, also known as Glioma-associated oncogene or oncogene GLI, from human origin. Anti-GLI-1 antibody (C-1) is raised against amino acids 781-1080 of the full-length GLI-1 protein, enabling effective detection in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). GLI-1 is a member of the GLI family of zinc finger transcription factors and plays a pivotal role in the Hedgehog signaling pathway, which is critical for embryonic development, tissue patterning, and the maintenance of stem cell populations. Located primarily in the nucleus, GLI-1 functions as a transcriptional activator by binding to specific DNA sequences and regulating the expression of target genes involved in cell proliferation, differentiation, and survival. Nuclear localization of GLI-1 is essential for modulating gene expression, and dysregulation is implicated in various cancers, including gliomas, highlighting GLI-1′s importance in both normal physiology and oncogenic processes. GLI-1 interacts with other proteins such as SUFU and various co-activators to finely tune transcriptional activity, ensuring precise control of cellular responses to developmental and environmental cues. Anti-GLI-1 antibody (C-1) is an indispensable tool for researchers studying the mechanisms of Hedgehog signaling, cancer biology, and developmental biology, providing valuable insights into the regulation and function of this critical transcription factor. Has it been tested for human IHC(P) yet? If it isn't, is there any Santa Cruz anti-Gli1 validated for IHC(P)? I have searched for the antibody you recommended two years ago but it seems it's not available anymore. Thank you very much

Thank you for your question. We have not yet tested this antibody on formalin-fixed paraffin-embedded tissue sections and so IHC(P) is not a recommended application at this time. We do currently offer an alternative antibody that has been tested for human IHC(P), GLI-1 Antibody (A-7): sc-515781.

I need to use an antibody to detect the protein in samples of avian origin. Do you know if sc-515751: GLI-1 (C-1) mouse monoclonal antibody will work?

Thank you for your question. We have not yet tested our, GLI-1 Antibody (C-1): sc-515751, on formalin-fixed paraffin-embedded tissue sections, so we do not know if it is suitable for IHC(P). We do currently warranty the antibody to work for IF, or immunohistochemistry or IF on fresh/frozen tissues/cells fixed in methanol or acetone. We do offer an alternative antibody to GLI-1 that is validated to work for IHC(P) on human tissues, GLI-1 Antibody (1B9F8): sc-517290 (https://www.scbt.com/scbt/product/gli-1-antibody-1b9f8)

Rated 5 out of 5 by from Best one we've used.We've used other Gli1 antibodies with mediocre results- inconsistent banding, short degradation time of the antibody, poor signal. This Gli1, with the corresponding m-IgGk BP-HRP secondary gave us great and repeatable Western Blot results on a sometimes tricky project. See the basic WB data below where a tagged Gli1 was over expressed in a cell line (first 3 lanes) and empty vector controls (last 3 lines).
