GLI-1 Antibody (A-7) is a mouse monoclonal IgG1 kappa light chain antibody engineered to recognize GLI-1, also known as Glioma-associated oncogene or oncogene GLI, from human origin. GLI-1 monoclonal antibody (A-7) is raised against a peptide spanning amino acids 1050-1068 near the C-terminus of the full-length GLI-1 protein, enabling detection in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). GLI-1 is a member of the GLI family of C2H2-type zinc finger transcription factors and plays a critical role in the Hedgehog signaling pathway, which is essential for embryonic development, tissue patterning, and the maintenance of stem cell populations. Structurally, GLI-1 contains multiple zinc finger domains that facilitate DNA binding and interaction with other proteins, such as SUFU and various co-activators, which regulate transcriptional activity. Post-translational modifications, including phosphorylation, are crucial for modulating GLI-1′s activity and stability, thereby influencing the ability to regulate target gene expression involved in cell proliferation, differentiation, and survival. GLI-1 is predominantly localized in the nucleus, where GLI-1 functions as a transcriptional activator, controlling the expression of genes that drive critical cellular processes and contribute to oncogenic transformation when dysregulated. The proper structural configuration of GLI-1 is essential for interaction with DNA and regulatory proteins, ensuring precise control of gene expression. Anti-GLI-1 antibody (A-7) is available in both unconjugated and conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, offering versatile options for a wide range of experimental applications. GLI-1 (A-7) antibody is an indispensable tool for researchers investigating the mechanisms of Hedgehog signaling, cancer biology, and developmental processes, providing valuable insights into the regulation and function of this pivotal transcription factor.
