Gem Antibody (A-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Gem protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Gem antibody (A-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Gem is a member of the Rad/Gem/Kir (RGK) subfamily of Ras-related GTPases, notable for its unique structure that lacks the typical C-terminal amino acid motifs required for isoprenylation, which is crucial for membrane localization and function. This structural characteristic allows Gem to interact with calmodulin in a calcium-dependent manner, specifically through residues 278-297 in human Rad, facilitating its role in cellular signaling pathways. High intracellular levels of Gem interact with microtubules and microfilaments, leading to significant alterations in cell morphology, such as cell flattening and neurite extension in neuroblastoma cell lines N1E-115 and SH-SY5Y, indicating involvement in cytoskeletal rearrangement and neuronal differentiation. Gem expression has been observed during the development of trigeminal nerve ganglia in 12.5-day mouse embryos, highlighting importance in normal ganglionic development. Gem interacts with beta-subunits to regulate the expression of calcium channels at the cell surface, underscoring a critical role in calcium signaling. The human Gem gene is located on chromosome 8q22.1, making Gem a significant target for studies related to neurodevelopment and cellular morphology.
