GCN2 Antibody (F-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects GCN2 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). GCN2 Antibody (F-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GCN2, also known as EIF2AK4, is a critical component of the cellular stress response, functioning as a kinase that phosphorylates the eukaryotic translation initiation factor 2α (eIF2α) on Ser 51. This phosphorylation event inhibits the guanine nucleotide exchange factor eIF2B, thereby regulating protein synthesis under stress conditions. GCN2 is particularly responsive to amino acid deprivation and UV radiation, making GCN2 essential for cellular adaptation to environmental stressors. GCN2 is expressed at high levels in liver and brain tissues, highlighting GCN2′s importance in metabolic regulation and neuronal function. The unique structure of GCN2 allows sensing of uncharged tRNA levels, which is crucial for maintaining cellular homeostasis during nutrient scarcity. The interaction of GCN2 with other stress-responsive kinases, such as HRI, PKR, and PERK, underscores GCN2′s integral role in the broader network of stress signaling pathways.

For the antibody sc-374609 , can I use Pfa 4% to fixe my tissue instead of methanol?

Thank you for your question. This antibody sc-374609 is recommended for detection of GCN2 of human origin by Western Blot, IP, Immunocytochemistry on methanol-fixed cells, Immunohistochemistry on acetone-fixed frozen tissue sections, Immunohistochemistry on paraffin-embedded tissue sections fixed with formalin and ELISA. We currently have no data for this antibody for a fixation with pfa. Please contact our Technical Service for additional information.

What is the recommended starting dilution for Western blot for GCN2 (F-7): sc-374609?
