GCAP2 Antibody (B-8) is a mouse monoclonal IgG2a antibody that detects guanylate cyclase-activating protein 2 (GCAP2) in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). GCAP2 plays a crucial role in the regulation of phototransduction in the retina, specifically in the outer segments of photoreceptors where GCAP2 is primarily located. GCAP2 is essential for the recovery of the dark state in rod photoreceptors after exposure to light, as GCAP2 modulates the synthesis of cyclic guanosine monophosphate (cGMP) in response to calcium levels. The unique feature of GCAP2, in contrast to other calcium-binding proteins, is GCAP2′s ability to activate guanylate cyclase in its calcium-free form, which is vital for maintaining photoreceptor function and preventing photoreceptor degeneration. GCAP2 is closely related to GCAP1, with both proteins sharing over 90% sequence similarity and three conserved calcium-binding sites, highlighting their evolutionary significance and functional redundancy in the retina. The genes encoding GCAP1 and GCAP2 are located on human chromosome 6p21.1, and GCAP2′s specific interactions with other proteins in the phototransduction cascade further underscore its importance in visual signaling pathways.
