Ganglioside GD2 Antibody (14G2a) is a mouse monoclonal IgG2a kappa light chain antibody that detects Ganglioside GD2 protein of human origin by immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Anti-Ganglioside GD2 antibody (14G2a) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Ganglioside GD2 is a sialic acid-containing glycosphingolipid that plays a crucial role in cell signaling and recognition, making Ganglioside GD2 essential for various cellular processes. Ganglioside GD2 presence on the cell surface facilitates interactions between cells and their environment, influencing cell adhesion and communication. Ganglioside GD2 is highly expressed in neuroblastoma cells and other tumors derived from neuroectoderm, such as malignant melanoma and certain gastrointestinal cancers, which underscores Ganglioside GD2′s potential as a target for immunotherapy. GM2/GD2 Synthase regulates Ganglioside GD2 production, and Ganglioside GD2′s involvement in apoptosis regulation through focal adhesion kinase dephosphorylation highlights Ganglioside GD2′s importance in cancer biology. Ganglioside GD2′s unique expression pattern on mesenchymal stromal cells suggests potential use as a distinctive surface marker, emphasizing relevance in both diagnostic and therapeutic applications.

Thank you for your question. This antibody was raised against neuroectodermal tumor cells of human origin. The exact epitope has not been epitope mapped. We are not able to provide the hybridoma for sequencing.

Dose Ganglioside GD2 (14G2a) has certificated of anaylsis for QC department for validation use?

From the description of the product, I understand that the 14(G2a) antibody is provided as the kappa light chain of the antibody. How can the kappa light chain recognise the GD2 sugar without the heavy chain? Many thanks

Thank you for the question. The Ganglioside GD2 (14G2a) is a full mouse IgG2a antibody. We characterized the type of light chain because instead of conventional secondary antibodies against full-IgG we instead use a light-chain-specific binding protein which tends to produce cleaner results compared to conventional secondary antibodies. In this case we suggest using m-IgGκappa-specific BP-HRP: sc-516102 for the detection of Ganglioside GD2 (14G2a) sc-53831.

How can this mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

When attempting a double or triple stain with mouse monoclonal antibodies, we recommend using a directly conjugated primary antibody. This product, sc-53831, is available conjugated to FITC, AF488, and AF647.
