FOXP2 Antibody (5C11A8) is a mouse monoclonal IgG1 kappa light chain antibody that detects Forkhead box protein P2 (FOXP2) protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-FOXP2 antibody (5C11A8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FOXP2 protein is a crucial transcription factor that plays a significant role in the development of neural circuits involved in speech and language, making FOXP2 essential for effective communication in humans. Mutations in FOXP2 gene have been linked to language and speech disorders, highlighting FOXP2′s importance in cognitive development and social interaction. FOXP2 protein is primarily located in the nucleus, where FOXP2 binds to specific DNA sequences to regulate the expression of target genes, thereby influencing neuronal development and function. Understanding FOXP2′s role in the brain can provide insights into the molecular mechanisms underlying language acquisition and the evolution of human communication. FOXP2 gene is located on chromosome 7q31, and FOXP2 expression is tightly regulated during development, particularly in areas of the brain associated with language processing.

Is there a recommended protocol for IF/IHC using this FoxP2 antibody (5C11A8)? I tried in rat tissue at 1:200, with negative results. I did not use an antigen retrieval procedure.

Thank you for your question. We do not currently recommend this FOXP2 Antibody (5C11A8): sc-517261 for IF or IHC(P), because we have not yet tested this antibody in either application. We also do not recommend this antibody for detection of FOXP2 of rat origin, because it has not yet been tested for reactivity in that species. This antibody is recommended for detection of FOXP2 of human origin by WB, IP and ELISA. In general, we would recommend using a starting dilution of 1:50 for IF/IHC, and if you are using formalin-fixation, then antigen retrieval is strongly advised.

Is this antibody raised against the same amino acids as the discontinued N-16 antibody?

Thank you for your question. FOXP2 Antibody (5C11A8): sc-517261 is raised against amino acids 47-287 of FOXP2 of human origin. This is a different epitope than the now discontinued goat polyclonal, FOXP2 Antibody (N-16): sc-21069, that was raised against a short peptide with an epitope mapping within the first 50 amino acids of the N-terminus of FOXP2 of human origin.
