EAAT2 Antibody (E-1) is a mouse monoclonal IgG2b kappa light chain antibody that detects EAAT2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). EAAT2 (E-1) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. EAAT2 protein, also known as solute carrier family 1 member 2 (SLC1A2) or glutamate transporter 1 (GLT-1), plays a crucial role in the central nervous system by mediating the uptake of the excitatory neurotransmitter L-glutamate, which is vital for maintaining synaptic homeostasis and preventing excitotoxicity. EAAT2 protein is predominantly localized in astrocytes and is essential for clearing excess glutamate from the synaptic cleft following neurotransmission, thereby protecting neurons from potential damage caused by glutamate accumulation. Proper functioning of EAAT2 is critical for normal brain function, as dysregulation of glutamate transport can lead to various neurological disorders, including epilepsy, neurodegenerative diseases, and mood disorders. EAAT2′s expression is primarily restricted to the brain, distinguishing EAAT2 from other glutamate transporters like EAAT1 and EAAT3, which are also found in non-nervous tissues. anti-EAAT2 antibody (E-1) allows researchers to explore the intricate mechanisms of glutamate transport and its implications in health and disease.

Is it possible to use this antibody with flow cytometry? is it against to an extracellular epitope?

Thank you for your question. The conjugated primary antibody is recommended and warrantied for flow cytometry (FCM), however we are still gathering data to show on our website. Check back for updates! This monoclonal antibody is raised against amino acids 1-85 mapping near the N-terminus of EAAT2, which encompasses potentially both cytoplasmic (aa 1-44) and extracellular (65-87) domains. The exact epitope recognized by this mouse monoclonal antibody has not yet been determined.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with EAAT2 Antibody (E-1): sc-365634?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
