Dnmt3a Antibody (C-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects Dnmt3a protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). anti-Dnmt3a antibody (C-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Dnmt3a protein plays a crucial role in regulating gene expression through its function as a DNA methyltransferase, specifically catalyzing methyl group addition to the 5′-position of cytosines within CpG dinucleotide context. This methylation maintains genomic stability and regulates transcriptional repression, particularly important during embryonic development and cellular differentiation. Dnmt3a, along with its closely related enzyme Dnmt3b, shows high expression in embryonic stem cells, where Dnmt3a establishes methylation patterns that dictate cell fate and lineage commitment. Dnmt3a′s ability to modify DNA highlights its significance in epigenetic regulation and potential implications in developmental biology and disease states, including cancer, where aberrant methylation patterns can lead to dysregulated gene expression.

Hi, I want to use this antibody for WB in tissue lysate from mouse hippocampus. Should I need to do nuclear cytoplasmic fraction? Or I can use RIPA buffer and use whole tisssue lysate?

Can I use the anti-Dnmt3a-AF647 antibody ( sc-365769 AF647) for flow cytometry? If so, do you have a recommended starting point for the concentration to be used?

Thank you for your question. sc-365769 AF647 has not been tested for flow cytometry in house, so we do not warranty it for that use, however it may work perfectly well. We recommend using ~1µg per million cells. I want to run a Western blot using this Dnmt3a antibody using NIH/3T3 cell lysate. How much should I load on my gel?
