DEP-1 Antibody (F-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects DEP-1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). DEP-1 (F-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Density-enhanced phosphatase-1 (DEP-1), also known as HPTP-eta/CD148, plays a crucial role in signal transduction within leukocytes and is integral to cellular differentiation processes. DEP-1 features a unique structure comprising an extracellular segment with eight fibronectin type III repeats, a single transmembrane segment, and an intracellular protein tyrosine phosphatase domain, which is essential for enzymatic activity. These fibronectin repeats facilitate protein-protein interactions and contribute to modulating signaling pathways. DEP-1 is predominantly expressed in lymphoid organs, including B and T cells, granulocytes, macrophages, and certain dendritic cells, as well as in non-lymphoid tissues such as fibrocytes, melanocytes, and various epithelial cell types with glandular or endocrine functions. In Jurkat T cells, DEP-1 inhibits T cell receptor-mediated activation, leading to decreased expression of early activation markers like CD69 and reduced tyrosine phosphorylation of key intracellular proteins, including the tyrosine kinase ZAP-70, thereby impairing mitogen-activated protein kinase activation. Despite phosphatase activity, DEP-1 can paradoxically induce protein tyrosine phosphorylation in human lymphocytes and serine/threonine or tyrosine phosphorylation in tumor cell lines, highlighting a complex regulatory role in cellular signaling.
