DEP-1 Antibody (143-41) is a mouse monoclonal IgG1 kappa light chain antibody that detects DEP-1 of human origin by western blotting (WB), immunoprecipitation (IP), and flow cytometry (FCM). DEP-1 (143-41) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Density-enhanced phosphatase-1 (DEP-1), also known as HPTP-eta/CD148, plays a crucial role in signal transduction within leukocytes and is integral to cellular differentiation processes. DEP-1 features an extracellular segment with eight fibronectin type III repeats, a single transmembrane segment, and a unique intracellular protein tyrosine phosphatase domain. This structural configuration allows DEP-1 to effectively interact with various signaling pathways, influencing immune cell activation and differentiation. In lymphoid organs, DEP-1 is prominently expressed on B and T cells, granulocytes, macrophages, certain dendritic cells, mature thymocytes, and neutrophils, while in non-lymphoid tissues, DEP-1 is found in fibrocytes, melanocytes, Schwann cells, and various epithelial cell types with glandular or endocrine functions. In Jurkat T cells, DEP-1 inhibits T cell receptor-mediated activation, leading to decreased expression of early activation markers such as CD69, reduced tyrosine phosphorylation of key intracellular proteins like ZAP-70, and impaired activation of mitogen-activated protein kinases. Despite intrinsic enzymatic activity, DEP-1 can paradoxically induce protein tyrosine phosphorylation in human lymphocytes and serine/threonine or tyrosine phosphorylation in certain tumor cell lines, highlighting a complex regulatory role in cellular signaling.

Can DEP-1 antibody 143-41:sc21761 be used to detect the 856-1313 aa fragment of tetanus toxoid protein? can this antibody conjugated to protein A?

Asked by: tetanus
Thank you for your inquiry. We do not characterize the binding sites of our monoclonal antibodies, so we cannot be any more specific about the reactivity of our DEP-1 (143-41) sc-21761 than that it was raised against PHA stimulated human PBL. For Special Orders including conjugation to Protein A, please contact our Special Orders team at specialorders@scbt.com
