Datasheets
  • CD3-ζ Antibody (E-3) is a mouse monoclonal IgG3 κ, cited in 8 publications, provided at 200 µg/ml
  • specific for an epitope mapping between amino acids 137-163 at the C-terminus of CD3-ζ of human origin
  • recommended for detection of CD3-ζ of mouse, rat and human origin by WB, IP, IF and ELISA; also reactive with additional species, including and canine, bovine and porcine
  • See CD3-ζ (6B10.2): sc-1239 for CD3-ζ antibody conjugates, including AC, HRP, FITC, PE, Alexa Fluor® 488, 594, 647, 680 and 790.
  • m-IgGκ BP-HRP is the preferred secondary detection reagent for CD3-ζ Antibody (E-3) for WB applications. This reagent is now offered in a bundle with CD3-ζ Antibody (E-3) (see ordering information below). For additional m-IgGκ BP conjugates see our complete list of Mouse IgG Binding Proteins.

SEE ALSO...

CD3-ζ Antibody (E-3) is a mouse monoclonal IgG3 kappa light chain antibody that detects CD3-ζ in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). CD3-ζ (E-3) antibody is available in a non-conjugated form, specifically designed for CD3-ζ detection. CD3-ζ plays a crucial role in T cell activation as a key component of the T cell receptor (TCR) complex, which is essential for recognizing foreign antigens. Upon antigen recognition, the TCR complex transmits signals that lead to T cell activation, proliferation, and differentiation, making CD3-ζ vital for the adaptive immune response. The structure of CD3-ζ is particularly important because CD3-ζ contains three immunoreceptor tyrosine-based activation motifs (ITAMs) that are critical for initiating signaling cascades upon TCR engagement. These ITAMs serve as docking sites for various protein kinases, including ZAP-70, which further propagate the activation signal within the T cell. The unique arrangement of ITAMs in CD3-ζ enhances signaling response amplification, ensuring a robust immune reaction. Understanding CD3-ζ function and structure is essential for developing therapeutic strategies aimed at modulating T cell responses in various diseases, including cancer and autoimmune disorders.

Can you recommend a negative control to use with CD3-ζ (E-3): sc-166435 monoclonal antibody?

Asked by: jenni
We recommend using the blocking peptide sc-166435 P as a negative control. Antibody binding to antigen may be blocked/competed by pre-absorption with the blocking peptide. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Is CD3-ζ (E-3): sc-166435 monoclonal antibody available directly conjugated to HRP or fluorescent dyes?

Asked by: jenni
Thank you for your question. All requests for directly conjugated versions of this antibody should be directed to our Special Orders team at specialorders@scbt.com Alternatively, consider using CD3-ζ (6B10.2): sc-1239, which is available conjugated to agarose, HRP, FITC, PE, and more. Rated 2 out of 5 by from Not ideal for immuno-staining HEK cellsI tried staining HEK293 cells that were transiently expressing a construct with the CD3 peptide and found that the signal was not very good compared to a parallel immuno-staining with another primary antibody

Rated 5 out of 5 by from good effectOur aim is to use T cell antigen receptor recognition of foreign antigens, and this recognition events into intracellular signals, causing cells from dormant to active state changes. This is a very good effect
