SEE ALSO...

CD3-ζ Antibody (6B10.2) is a mouse monoclonal IgG1 kappa light chain antibody that detects CD3-zeta of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). CD3-ζ (6B10.2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD3-zeta plays a crucial role in T cell activation and signaling, as CD3-zeta is a key component of the T cell receptor (TCR) complex, which is essential for the immune response. Located in the plasma membrane of T cells, CD3-zeta is part of a multisubunit complex that transmits signals upon recognition of foreign antigens by the TCR. This signaling cascade is vital for the activation of T cells, leading to their proliferation and differentiation into effector cells that can target and eliminate pathogens. The structure of CD3-zeta, which contains three immunoreceptor tyrosine-based activation motifs (ITAMs), allows for efficient recruitment of signaling molecules, such as ZAP-70, that further propagate the activation signal. The ability of CD3-zeta to form homodimers or heterodimers with other CD3 components enhances the complexity and specificity of T cell signaling, making CD3-zeta a critical target for research in immunology and therapeutic interventions.

In the description "raised against amino acids 36-54 mapping within an N-terminal domain of CD3-ζ of human origin" However, aa 36-54 is almost in the TM domain (31-51). How do you define aa 36-54? with or without leader? can it detect cytoplasmic one

Asked by: Leo_
Thank you for your question. The exact epitope recognized by this mouse monoclonal antibody has not yet been determined. We do have IHC(P) data on our website using this antibody to detect membrane and cytoplasmic staining in formalin-fixed paraffin-embedded tissue sections.

Is the agarose-conjugated CD3-ζ Antibody (6B10.2): sc-1239 provided as 500µg/mL, unlike the other formats that come as 200µg/mL? The recommended concentration for IP is 1-2µg/mL--does this mean that you would only need 2-4µL agarose-conj. Ab per 1mL IP?

Asked by: eleonard
Thank you for your question. For the agarose conjugated antibody, sc-1239 AC, we recommend using ~ 20uL of a 25% slurry, which equals 10 ug of ab/5 uL of agarose beads. Each vial contains 500 ug of antibody/250 uL of agarose in 1 mL of slurry (PBS + Sodium Azide + conjugated antibodies).

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with CD3-ζ Antibody (6B10.2): sc-1239?

Asked by: AbPolly
We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety
