AQP2 Antibody (E-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects AQP2 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-AQP2 antibody (E-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AQP2 plays a crucial role in regulating water balance in the body, specifically by facilitating water reabsorption in the kidney collecting ducts in response to vasopressin. This function is vital for maintaining homeostasis, particularly in conditions of dehydration or increased plasma osmolality. AQP2 is unique among aquaporins as the only water channel directly regulated by vasopressin, which enhances activity and promotes urine concentration. Proper functioning of AQP2 is essential for preventing conditions such as diabetes insipidus, where the body fails to concentrate urine, leading to excessive urination and thirst. Understanding AQP2 regulation is important for developing therapeutic strategies for kidney-related disorders and managing fluid balance in various clinical settings.

Hello, what is the recommended IHC protocol for this antibody? I would particular like to know about antigen retrieval, blocking and primary antibody incubation time/dilution.

We recommend antigen retrieval using citrate buffer (also described in the protocol) and our plant-based UltraCruz® Blocking Reagent: sc-516214 for blocking. How can AQP2 (E-2): sc-515770 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to HRP (sc-515770 HRP), phycoerythrin (sc-515770 PE), fluoroscein (sc-515770 FITC) and AlexaFluors.

Rated 5 out of 5 by from Work in kidney paraffin sectionsI tried this antibody on human baby (6 weeks) kidney paraffin sections for 1:25 dilution and it worked nicely. It was antigen retrieved with heat mediated antigen retrieval in 1mM EDTA buffer pH 8.0. It labeled the principal cells in the collecting duct as reported in literature.

Rated 5 out of 5 by from Works great on paraffin section immunostainingI tried this antibody on embryonic day E16.5 mouse kidney paraffin sections for 1:200 dilution and it worked nicely. The kidney was fixed in 4% PFA for 3 hours and sectioned at 10um thickness. It was antigen retrieved as well. It labeled the principal cells in the collecting duct as reported in literature.
