α/β-synuclein Antibody (Syn 202) is a mouse monoclonal IgG2a kappa light chain antibody engineered to detect both α-synuclein and β-synuclein proteins across human, mouse, and rat species. α/β-synuclein monoclonal antibody (Syn 202) targets an epitope on human recombinant α/β-synuclein, and is recommended for application in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry techniques. Synucleins, including α-synuclein (also known as NACP), β-synuclein (PNP 14), and γ-synuclein (persyn or BCSG1), are presynaptic proteins that play crucial roles in synaptic regulation and neuronal plasticity, primarily within the brain. α-Synuclein is notably a component of Alzheimer′s disease amyloid plaques and is localized to neuronal cell bodies and synapses, where post-translational modifications, such as phosphorylation and ubiquitination, are essential for function and implicated in the pathology of neurodegenerative diseases like Parkinson′s disease. These modifications influence protein aggregation and toxicity, making them critical targets for research and therapeutic strategies. β-Synuclein is involved in hematopoietic cell differentiation, while γ-synuclein is linked to axonal pathology in Parkinson′s disease, highlighting the diverse roles of synuclein family members in neuronal health and disease. α/β-synuclein (Syn 202) antibody is available in both unconjugated form and various conjugated forms, including horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing flexibility for different experimental requirements. By recognizing both native and modified forms of synucleins, α/β-synuclein (Syn 202) monoclonal antibody serves as a valuable tool for investigating the molecular mechanisms underlying synaptic function and neurodegenerative disorders.

What is the epitope recognised?

Asked by: chiara9227
Thank you for your question. The antibody was raised against recombinant human a/b synuclein. The recognition epitope is located within the aas 130-140 of a synuclein with protein accession number https://www.ncbi.nlm.nih.gov/protein/P37840 Please contact our Technical Service by phone or email or by Live Chat if you have any further questions.

I am using α/β-synuclein (Syn 202): sc-32281 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Cweed
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
