AHA-1 Antibody (F-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects AHA-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). AHA-1 (F-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AHA-1 plays a pivotal role in regulating heat shock protein 90 (HSP 90) complex, which is essential for protein folding and stabilization under stress conditions. Specifically, AHA-1 enhances HSP 90 ATPase activity, facilitating function in protein maturation and degradation pathways. Such regulation influences "ATP lid" conformational state and promotes N-terminal dimerization, crucial for HSP 90′s activity. AHA-1 predominantly resides in cytosol but can transiently associate with endoplasmic reticulum, impacting protein trafficking from endoplasmic reticulum to Golgi apparatus. Heat shock induces AHA-1 expression in various tissues, including brain, heart, skeletal muscle, and kidney, with lower levels in liver and placenta. AHA-1′s ability to maintain cell viability under non-optimal growth conditions highlights significance in cellular stress responses, making anti-AHA-1 antibody (F-7) valuable for studying protein homeostasis and stress signaling pathways.

Why is the band sometimes at 50 KD sometimes at 43 KD and sometimes not visible at all in YOUR own slides of the WB?

Thank you for your question. AHA-1 has a predicted molecular weight of 38 kDa. We typically observe this in the whole cell lysates tested at ~43 kDa when using our Cruz Marker Molecular Weight Standard (sc-2035). There is one published Western blot on our website where we instead used our Prestained Broad Range Markers (sc-2361) where the signal detected in the mouse AHA-1 transfected 293T whole cell lysate is observed slightly below the 50 kDa prestained marker, so close to 43 kDa. The difference in the observed molecular weight is due to the different molecular weight marker standards used. Please note that we do not always expect to detect any signal in the non-transfected 293T whole cell lysates. We have tested this AHA-1 antibody for specificity against different human and mouse AHA-1 transfected 293T whole cell lysates. In some cases, depending on the testing conditions (e.g., antibody/antigen concentration and exposure time) we may detect a signal in the non-tranfected control. However, the goal with the published data is to show reactivity with the overexpressed protein.

I need to use an antibody to detect the protein in samples of equine origin. Do you know if sc-166610: AHA-1 (F-7) mouse monoclonal antibody will work?

Rated 5 out of 5 by from WBできれいに検出できました抗体はWBに使用しました。サンプルはJurkatとMT-2細胞株。抗体の希釈倍率は1000倍。ブロッキング剤は市販のブロッキング剤または5%スキムミルクを使用しました。どちらのブロッキング剤を使用してもバックグラウンドはかなり低く、きれいなバンドが検出されました。
