ACTH/CLIP Antibody (G-11) is a mouse monoclonal IgG1 kappa light chain antibody that specifically binds to an epitope mapping between amino acids 158-183 within an internal region of Pro-opiomelanocortin (POMC), also known as corticotropin-lipotropin. POMC is a 267 amino acid polypeptide hormone precursor that undergoes extensive tissue-specific post-translational modifications by prohormone convertases, resulting in the production of multiple active peptides such as Adrenocorticotropic Hormone (ACTH) and Corticotropin-like Intermediate Peptide (CLIP). These post-translational modifications enable POMC to generate bioactive peptides that regulate essential physiological processes including stress response, metabolism, and pigmentation. Mutations or defects in the POMC gene or processing enzymes can lead to POMC deficiency, characterized by red hair, adrenal insufficiency, and increased susceptibility to obesity. ACTH, also known as corticotropin, is a 39 amino acid peptide that stimulates cortisol secretion from the adrenal cortex, playing a vital role in the body′s response to stress. CLIP is a 21 amino acid neuropeptide secreted by corticotrope cells of the adenohypophysis, involved in various neurological functions. POMC also gives rise to other peptides like melanocyte-stimulating hormones (α-MSH, β-MSH, γ-MSH), β-endorphin, and Met-enkephalin, which have roles in pigmentation, energy homeostasis, and pain modulation. POMC (G-11) antibody reacts with POMC and processed active peptides ACTH and CLIP from mouse, rat, and human origins, and is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) applications.
