Topo IIβ Antibody (A-12) is a mouse monoclonal IgG2a kappa light chain antibody that detects Topo IIβ in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Topo IIβ antibody (A-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Topo IIβ (A-12) monoclonal antibody recognizes a protein that plays a crucial role in maintaining DNA integrity during critical cellular processes such as replication and transcription by introducing transient double-strand breaks in DNA, allowing for relaxation of supercoiled DNA structures. This function prevents DNA tangling and ensures proper chromosome segregation during cell division, which is essential for genomic stability. Differential expression of Topo IIβ, particularly in specific tissues, suggests unique regulatory roles in cellular processes compared to Topo IIα isoform. Understanding distinct functions of Topo IIβ can provide insights into potential therapeutic targets in cancer treatment, where modulation of topoisomerase activity is a common strategy for disrupting cancer cell proliferation.

Blocking in milk for hour. Incubateing with first antibody(1:500) overnight. Incubateing with second antibody(1:2000) for one hour

Rated 2 out of 5 by from TOPOIIB A-12 in Western BlotI received this as a free sample antibody and tested it out at a 1:200 dilution using Whole Cell Extracts from four different cell lines (HeLa, HEK293, HEK293T, and Mouse Embryonic Fibroblast cells). The expected size for TOPO11B is 180 kDa and I do see something at the correct height in maybe the last 3 lanes (the HeLa could just be loaded with less extract). The exposure of the blots was weird for this antibody and another Topoisomerase antibody I also tested (Santa Cruz H-8).
