Rotavirus p42 is an integral protein of the rotavirus particle, a double-stranded RNA virus known to cause severe diarrheal illness in infants and young children globally. The p42 protein is part of the triple-layered protein capsid that encapsulates the viral genome.The p42 protein, also known as VP4, is a spike protein that protrudes from the outermost layer of the virus particle. It is synthesized as a precursor protein (VP4), which is then cleaved by a host enzyme into two proteins: VP8* and VP5*, collectively still referred to as VP4 or p42.
VP4 plays a crucial role in the life cycle of the rotavirus, mediating both virus attachment to host cells and membrane penetration for entry. The VP8* subunit is responsible for binding to specific receptors on the surface of host cells, while the VP5* subunit plays a role in virus penetration into the cytoplasm.The p42 protein is also a key target of the immune response. Antibodies against VP4 can neutralize the virus, blocking it from entering host cells. This makes it an important antigen in rotavirus vaccines.Furthermore, VP4 is classified into P genotypes based on its genetic variability.
