PDPR Activators

PDPR Activators consist of chemicals known to facilitate the operation of pyruvate dehydrogenase phosphatase and the pyruvate dehydrogenase complex, either directly or indirectly. These chemicals do not inhibit PDPR but rather necessitate or complement its activity. Compounds such as Coenzyme A and NAD+ specifically act to increase the rate or efficiency of the PDC, which in turn would require PDPR's regulatory influence. Calcium Chloride and Alpha-Ketoglutarate are unique in that they serve as cofactors or substrates within the cellular pathways linked to PDPR, thereby naturally creating a need for PDPR's unique modulatory action.

Other activators like Dichloroacetate (DCA) or Isoproterenol function through a more convoluted route. DCA inhibits PDK, the kinase that phosphorylates and inactivates PDC. The inhibition of PDK leads to an activation of PDC, thereby requiring PDPR for its regulatory action. Isoproterenol works by activating the cAMP pathway which can indirectly influence the activity of PDC, creating an environment where PDPR's role becomes crucial. All these chemicals can, therefore, be understood as serving a role in the upregulation of PDPR's modulatory functions.
