Neurogenin 2 Activators

Neurogenin 2 activators constitute a class of molecules that have been shown to induce expression of the neurogenin 2 protein. These activators function through a variety of intracellular pathways to enact changes in the cell that enhance neurogenin 2 gene transcription. Many activators activate gene transcription cascades such as the cAMP/PKA, MAPK/ERK, Wnt/β-catenin, and calcium/calmodulin pathways through receptor binding or other upstream mechanisms.

Activation of these cellular signaling modules leads to a series of phosphorylation events that indirectly or directly influence the activity of neurogenin 2 transcription regulators. For instance, phosphorylation may modify the DNA binding affinity or protein-protein interaction abilities of transcription factors that occupy neurogenin 2 promoter or enhancer regions. Alternatively, phosphorylation targets like histone proteins or transcriptional coactivators could mediate enhanced recruitment of the transcriptional machinery. Some neurogenin 2 activators also hamper the function of transcription inhibitory complexes or DNA methyltransferases, thereby reducing genomic repression of the neurogenin 2 locus. By modulating multiple intracellular transduction networks and the phosphorylation state of diverse transcription regulatory components, neurogenin 2 activators demonstrate diverse tactics for overcoming pathways that repress baseline neurogenin 2 gene expression. This in turn loosens transcriptional control and augments neurogenin 2 mRNA production. Elucidating the mechanisms by which individual activators function within these modulatory cascades continues to advance insight into natural modes of neurogenin 2 transcriptional governance.
