Jagged1 Activators

Jagged1, a type I transmembrane protein, serves as a critical ligand in the Notch signaling pathway, a highly conserved signaling cascade that regulates various cellular processes during development and homeostasis. The function of Jagged1 primarily involves mediating cell-cell communication by interacting with Notch receptors on adjacent cells. Upon binding to Notch receptors, Jagged1 initiates a series of proteolytic cleavage events that ultimately lead to the release of the Notch intracellular domain (NICD). This liberated NICD translocates into the nucleus, where it forms a transcriptional activation complex with other proteins, such as CSL (CBF1/Su(H)/Lag-1) and Mastermind-like (MAML), thereby regulating the expression of target genes involved in cell fate determination, differentiation, proliferation, and apoptosis.

The activation of Jagged1-mediated Notch signaling occurs through a well-coordinated process involving ligand-receptor interactions and subsequent intracellular signaling events. Initially, Jagged1 binds to the extracellular domain of Notch receptors, including Notch1-4, through specific protein-protein interactions. This interaction induces conformational changes in the Notch receptor, leading to the exposure of cleavage sites susceptible to proteolytic processing by enzymes such as ADAM metalloproteases. Subsequently, the transmembrane domain of Notch is cleaved by γ-secretase, releasing the NICD into the cytoplasm. Upon NICD release, it translocates to the nucleus and interacts with transcriptional regulators to modulate gene expression. The activation of Jagged1 thus serves as a crucial step in initiating Notch signaling, orchestrating diverse cellular responses essential for embryonic development, tissue homeostasis, and disease pathogenesis.
