Shank 1/2/3 Antibody (G-12) is a mouse monoclonal IgG2a kappa light chain antibody that detects Shank 1/2/3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Shank 1/2/3 (G-12) monoclonal antibody is specific for an epitope mapping between amino acids 2124-2153 at the C-terminus of Shank 1 of human origin. Anti-Shank 1/2/3 antibody (G-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Shank proteins, which are part of the Shank/ProSAP family, serve as crucial molecular scaffolds within the postsynaptic density (PSD), a specialized structure that plays a vital role in synaptic transmission. PSD is located beneath the postsynaptic membrane of excitatory synapses and is essential for anchoring and clustering glutamate receptors, thereby facilitating effective neurotransmission. Shank proteins are characterized by PDZ domains, which are instrumental in mediating protein-protein interactions by recognizing specific sequence motifs at the C-terminus of target proteins. This interaction is critical for synaptic localization of various ion channels, receptors, signaling enzymes, and cell adhesion molecules. Shank proteins recruit betaPIX and PAK to dendritic spines, influencing structural integrity of postsynaptic sites and interacting with NMDA and metabotropic glutamate receptor complexes. Transcript splice variants within the Shank family allow for a dynamic spectrum of Shank-interacting proteins in PSDs of both developing and adult brains, ensuring proper synaptic function and development.
