VARS2 Activators

Chemical activators of VARS2 can engage multiple cellular signaling pathways to enhance the protein's function. Insulin, for instance, activates the PI3K/AKT signaling pathway, leading to the phosphorylation of various proteins, including aminoacyl-tRNA synthetases like VARS2. This post-translational modification can increase protein synthesis, thereby enhancing the activity of VARS2 in its role in tRNA aminoacylation. The activation of the MAPK/ERK pathway by Epidermal Growth Factor (EGF) also contributes to the phosphorylation of translation machinery components, which may include VARS2, thus activating its tRNA-charging function.

Additional activators like L-Leucine and Resveratrol exert their effects through mTOR and SIRT1 pathways respectively. L-Leucine can activate mTOR, which is a central regulator of cell metabolism, growth, proliferation, and survival, and can enhance the phosphorylation state of aminoacyl-tRNA synthetases, thus potentially increasing VARS2 activity. Resveratrol, a known activator of SIRT1, can deacetylate and activate components of the protein synthesis apparatus, which may include VARS2, leading to increased functional activity. Metformin and AICAR, both activators of AMPK, can modulate the cellular energy state and have downstream effects on the translation machinery, which can result in the enhanced activity of VARS2. GW501516, a PPARδ agonist, can increase the expression of proteins involved in energy metabolism, possibly providing the cellular energy required for VARS2 activation. Rapamycin, though known to inhibit mTOR, also indirectly activates autophagy, thus potentially increasing the functional availability of VARS2 by the degradation of misfolded proteins. Similarly, Spermidine induces autophagy, which can lead to the removal of dysfunctional cellular components and enhance the activation of VARS2. Lastly, Lithium can activate the Wnt/β-catenin pathway, implicated in regulating protein synthesis, which can lead to increased activation of VARS2 in its essential role in the aminoacylation of tRNA.
