TRPT1 Inhibitors

Chemical inhibitors of TRPT1 function by impacting the availability of its substrate, uric acid, which TRPT1 is known to process. Allopurinol, Febuxostat, Topiroxostat, Oxypurinol, and Tisopurine operate through a shared mechanism, inhibiting xanthine oxidase, an enzyme responsible for the conversion of hypoxanthine and xanthine to uric acid. By inhibiting xanthine oxidase, these chemicals reduce the synthesis of uric acid, thereby limiting the amount available for TRPT1 activity. Similarly, Rasburicase and Pegloticase directly convert uric acid into allantoin, a more soluble and easily excretable compound. This conversion results in a decline in uric acid levels, which in turn diminishes the substrate that TRPT1 would typically act upon.

In a complementary fashion, Benzbromarone, Probenecid, Sulfinpyrazone, and Lesinurad facilitate the excretion of uric acid from the body. Benzbromarone and Sulfinpyrazone function as uricosuric agents, which enhance the renal excretion of uric acid, thereby reducing its plasma concentration and the amount available for TRPT1. Probenecid also increases renal excretion of uric acid, further contributing to a decrease in available substrate for TRPT1. Lesinurad inhibits uric acid transporter proteins, which results in decreased reabsorption and increased clearance of uric acid from the renal system, again limiting the uric acid pool for TRPT1. Lastly, while Guaifenesin is primarily recognized for its expectorant capabilities, it can also influence urate solubilization, which may lead to reduced uric acid levels, indirectly limiting the substrate for TRPT1.
