Trmt2b Inhibitors

Chemicals classified as TRMT2B inhibitors encompass a variety of compounds that interact with the methylation process of RNA. These compounds can either mimic the structure of the natural substrates of methyltransferases or compete with them, thereby inhibiting their function. Adenosine analogs such as Adenosine Periodate, 3-Deazaneplanocin A, and Neplanocin A can disrupt the normal function of tRNA methyltransferases by interfering with the adenosine residues or the methylation cycle. Sinefungin and Methylthioadenosine are also adenosine analogs that inhibit methyltransferases by occupying the SAM binding site, which is critical for the transfer of methyl groups to tRNA. Other compounds, like RG108, AZA, and Decitabine, are nucleoside analogs that can be incorporated into RNA molecules during transcription. Their incorporation may alter the structure of tRNA, making it a less suitable substrate for TRMT2B or directly inhibiting the enzyme's activity. BIX-01294 and Furamidine, though primarily known for their inhibitory effects on other types of methyltransferases, could exhibit cross-reactivity with TRMT2B due to structural similarities between these enzymes. Amiloride and Hydralazine have been noted to affect RNA and DNA methylation, respectively. Their inclusion in this list is based on the premise that their action on nucleic acid methylation can extend to tRNA methylation, potentially affecting TRMT2B's activity.
