Chemical inhibitors of TRIM40 can exert their effects through the inhibition of the proteasome, a complex that degrades ubiquitinated proteins. As TRIM40 is implicated in tagging proteins for degradation, the use of proteasome inhibitors can lead to an accumulation of these proteins, effectively inhibiting the function of TRIM40. MG132, Lactacystin, Epoxomicin, Bortezomib, Carfilzomib, Ixazomib, Oprozomib, Delanzomib, Marizomib, Nelfinavir, Withaferin A, and Celastrol are all chemicals that inhibit the proteasome and can therefore impact the activity of TRIM40. For instance, MG132 can prevent the degradation of ubiquitinated proteins, disrupting the cellular pathway in which TRIM40 participates, causing an increase in proteins that TRIM40 might otherwise target for degradation. Similarly, Lactacystin acts on the proteasome to prevent it from breaking down proteins, which could include substrates of TRIM40.
Bortezomib and Carfilzomib are known to bind to the proteasome reversibly and irreversibly, respectively, impeding the proteolytic function that would normally regulate the turnover of proteins involved in TRIM40's activity. Ixazomib and Oprozomib, through their proteasome inhibitory action, can lead to an increased half-life of proteins that TRIM40 tags for degradation. Delanzomib and Marizomib, by inhibiting the proteasome, can alter the degradation pathway of proteins that are under the regulatory influence of TRIM40. Nelfinavir's inhibition of the proteasome can also lead to an accumulation of ubiquitinated proteins, indirectly inhibiting TRIM40's function. Withaferin A and Celastrol, which inhibit proteasomal activity, can similarly affect the turnover of proteins and disrupt the degradation process that TRIM40 is designed to facilitate. These chemicals, by inhibiting the proteasome, impair the degradation pathway of the ubiquitin-proteasome system, affecting the regulation and function of TRIM40.
