THOC2 Activators

THO complex subunit 2, often abbreviated as THOC2, is a protein that in humans is encoded by the THOC2 gene. This protein is a component of the THO complex, which is a key constituent of the larger transcription export (TREX) complex. The TREX complex has a critical role in the coupling of mRNA processing to mRNA export from the nucleus to the cytoplasm.The THO complex itself, including THOC2, is involved in the process of transcription elongation within the nucleus of the cell. It helps in the maintenance of genome integrity during mRNA synthesis by inhibition the formation of RNA-DNA hybrids known as R-loops, which can occur if nascent RNA re-hybridizes with the DNA template strand. Such structures can lead to genomic instability if not properly managed.

THOC2, like other components of the THO complex, is involved in ensuring efficient RNA export, particularly for mRNA molecules that are long or have complex secondary structures that could impede their movement through the nuclear pore complex. It achieves this by facilitating the proper packaging of mRNA and its association with export factors.Mutations in the THOC2 gene have been associated with various forms of intellectual disability, suggesting its crucial role in the development and function of the nervous system. Moreover, because of its fundamental role in gene expression and maintenance of genomic stability, THOC2 and other components of the TREX complex are also subjects of interest in the study of cancer and other diseases where these processes are disrupted.
