TFIIE-β Inhibitors

TFIIE-β inhibitors constitute a class of chemical compounds that primarily target the transcription factor IIE-beta (TFIIE-β) within the context of transcriptional regulation. TFIIE-β is a crucial component of the transcription preinitiation complex responsible for the accurate initiation of RNA synthesis by RNA polymerase II. The function of TFIIE-β revolves around its interaction with other transcription factors and RNA polymerase II, playing a pivotal role in the assembly of the transcription initiation machinery. Inhibition of TFIIE-β is characterized by the disruption of these critical interactions, leading to a cascade of effects that hinder transcriptional initiation.

The mechanisms of action of TFIIE-β inhibitors can be diverse. Some inhibitors, like triptolide and curcumin, directly bind to the active site of TFIIE-β, interfering with its ability to interact with other components of the transcription initiation complex. This disruption impedes the assembly of the preinitiation complex and consequently inhibits the initiation of transcription. Others, such as flavopiridol and α-amanitin, target downstream elements of the transcription machinery. Flavopiridol, for example, inhibits the kinase activity of TFIIE-β, preventing the phosphorylation of RNA polymerase II and effectively stalling transcription. α-Amanitin, on the other hand, binds directly to RNA polymerase II, preventing its dissociation from the DNA template. This interaction halts transcription by physically obstructing the progression of the polymerase along the DNA strand. TFIIE-β inhibitors represent a diverse class of compounds that exert their inhibitory effects through various mechanisms.
